Public and private lab price: 82.24 €/hour

Brief description:
Nikon widefield inverted microscope for epifluorescence and TIRF acquisition for 2D and 3D live imaging coupled to the FRAP/photo-activation/photoconversion technics. This microscope is also dedicated to the super-resolution technics (2D-3D) as PALM and STORM.
Controlled by Metamorph software
Objectives:
10X Plan Apochromat 0.45NA air
20X Plan Apochromat 0.75NA air
60X Plan Apochromat 1.4NA oil
100X Plan TIRF Apochromat 1.49NA oil

Detectors:
1 EMCCD iXon 897 Ultra Andor camera
(512*512, 16µm pixel size)

Incubation:
Temperature and CO2 control.
Acquisition thanks to the Roper iLAS2:
-widefield microscopy
-multi-colour TIRF microscopy
-FRAP/photo-activation/photo-conversion in single point, predefined ROI or on the fly
-PALM or STORM super-resolution

The Multi-dimensional Acquisition module in Metamorph controls the multi-series of images in multi-channels, multi-positions, multi-Z planes, multi-timepoints.
Illumination sources:
1 Lumencor with 7 LED and excitation filters for:
DAPI, CFP, GFP, YFP, Cy3, Cherry, Cy5
1 laser bench with:
405nm (100mW), 488nm (150mW), 561nm (500mW) et 642nm (1W)

Spectral specificities:
-EMCCD camera: Hoechst, GFP, mCherry, Cy5 with laser excitation or GFP and Cherry with LED excitation
-sCMOS camera: Hoeschst, CFP, GFP, Cy3, mCherry with LED excitation
Compatible sample carriers:
35mm dish with #1.5 glass bottom, glass slide with #1.5 glass coverslip or glass-bottom multi-well plate
