Laser ablation
Bruker upright microscope coupled with two Coherent lasers, enabling cellular and tissue imaging of live or fixed samples, and integrated with electrophysiology. Prairie View acquisition software.
Objective: 20X Olympus XLUMPLFLN (N.A. 1) water immersion, WD 2 mm.
Detectors: 3 GaAsP NDD detectors.
Acquisition modes: XYZ multi-photon confocal imaging, time-course, multi-position or mosaic. Photostimulation/photoactivation experiments. The system is equipped with a galvanometric scanner and a resonant scanner. Multi-photon imaging can be coupled with electrophysiological recordings.
Customisable protocols.
Excitation sources: Chameleon Discovery NX (Coherent) dual-line laser: one tunable line in the 660–1320 nm range and one fixed line at 1040 nm. A delay line enables the temporal synchronisation of the tunable and fixed laser beams.
Spectral configuration:
For two-photon imaging, an LP 565 dichroic filter combined with BP 525/70 and BP 595/50 filters, and an LP 495 dichroic filter combined with a BP 485/25 filter.
Full-field observation: fluorescence and transmission. A GFP cube (470/40, 495LPXR, 525/50) and a DsRed cube (545/30, 570LP, 620/60) combined with a Retiga Electro CCD camera for observation and navigation across the sample.
Sample holders: Slice Mini bathchamber II (Luigs&Neumann).
